Thyroxine-Induced Cardiac Hypertrophy and Adrenoceptor Changes

چکیده

Short-term (5 days) administration of L-thyroxine (0.1 mg/kg i.p. daily) to adult SpragueDawley rats induces a modest degree of cardiac hypertrophy (22% increase in heart weight/body weight ratios) and directionally opposite changes in cardiac adrenoceptors (24% increase in fi-adrenoceptors and 201% decline in ae-adrenoceptors). Pretreatment with colchicine did not affect the ability of L-thyroxine to induce cardiac hypertrophy but prevented its effects on both ffand ael-adrenoceptors. These results suggest a selective involvement of microtubules in the action of thyroid hormones on the expression of cardiac adrenoceptor genes. (Circulation Research 1991;68:309-313)

Protein synthesis and degradation during regression of thyroxine-induced cardiac hypertrophy.

To characterize changes in rates of protein turnover during regression of thyroxine-induced left ventricular hypertrophy, New Zealand White rabbits received intravenous thyroxine (200 micrograms/kg/d) for 9 days. Thyroxine was withheld, and in vivo protein turnover was evaluated on the 10th, 15th and 20th days. Animals not receiving thyroxine served as controls. Heart rate, blood pressure, and ...

Synthesis and degradation of myocardial protein during the development and regression of thyroxine-induced cardiac hypertrophy in rats.

Cardiac hypertrophy was induced in rats by daily injections of L-thyroxine (1.0 mg/kg). Regression from hypertrophy was studied 4 days after discontinuing thyroxine. Isolated, Langendorff-perfused hearts were perfused with Krebs-Henseleit buffer, glucose, insulin, and amino acids. To measure protein synthesis, left ventricular tissue was assayed for incorporation of tritiated phenylalanine into...

Dynamic changes of hemodynamic parameters and cardiac transcription of sirtuins in adaptive and mal-adaptive phases of pressure overload-induced hypertrophy in rats

Introduction: The aim of the study was to investigate the structural and hemodynamic changes as well as cardiac transcriptional profile of the key regulatory proteins, sirtuins family (SIRT1-7), in adaptive and mal-adaptive phases of left ventricular hypertrophy (LVH). Methods: LVH was induced in male Wistar rats (190±20g) by abdominal aortic banding. The third and sixteenth weeks post-surgery ...

The Possible Role of TNF-alpha in Physiological and Pathophysiological Cardiac Hypertrophy in Rats

Pathological cardiac hypertrophy was produced by partial abdominal aortic constriction (PAAC) for 4 wk, while physiological cardiac hypertrophy was produced by chronic swimming training (CST) for 8 wk in rats. Pentoxifylline (30 mg/kg, 300 mg/kg i.p., day-1) treatment was started three days before PAAC and CST and it was continued for 4 wk in PAAC and 8 wk in CST experimental model. The left ve...

Characterization of cardiac hypertrophy and heart failure due to volume overload in the rat.

Alterations in general characteristics and morphology of the heart, as well as changes in hemodynamics, myosin heavy chain isoforms, and beta-adrenoceptor responsiveness, were determined in Sprague-Dawley rats at 1, 2, 4, 8, and 16 wk after aortocaval fistula (shunt) was induced by the needle technique. Three stages of cardiac hypertrophy due to volume overload were recognized during the 16-wk ...
